Role Overview
We are seeking a passionate Research Associate II/Senior RA to support upstream production and process development of TCR bispecifics and MHC proteins in the Protein Engineering and Molecular Sciences team. The ideal candidate will have hands-on experience working with mammalian and/or E. coli cell culture, with preferred experience extracting inclusion bodies from E. coli. The incumbent will work at the bench to develop production cell lines, establish and maintain working cell banks and RCBs, and optimize cell culture conditions for TCR bispecifics and MHC proteins, as well as extraction methods for inclusion bodies. The candidate will be responsible for upstream production and process development, data analysis and presenting data to their supervisor and to the team in an effective and timely manner. Interested candidates should have strong organizational, interpersonal, verbal, and written communication skills that will enable them to integrate with our highly cross-functional team.

Key Responsibilities
  • Test plasmid/host combinations for optimal E. coli and mammalian expression.
  • Establish and maintain working cell banks and RCBs.
  • Optimize cell culture conditions to increase yield (E. coli and mammalian).
  • Develop robust processes for inclusion body extraction and mammalian supernatant clarification.
  • Execute multi-liter upstream runs to produce inclusion bodies and clarified supernatants for transfer to downstream processing.
  • Characterize recombinant protein expression using SDS-PAGE, Western Blot and Octet.
  • Capture primary data, establish and maintain SOPs, and document work as required in Electronic Laboratory Notebook (ELN), maintaining up to date records.
  • Present data routinely to the Protein Sciences group and other multidisciplinary groups.

Qualifications/Experience
  • BS/MS in Biochemistry, Biology, Bioengineering, Immunology, or a similar field.
  • 2+ years of academic or biotech/pharma laboratory experience.
  • Experience with recombinant protein expression using mammalian cell lines and/or E. coli.
  • Experience with inclusion body extraction/purification a plus.
  • Strong organizational and communication skills along with excellent attention to details.
  • Enjoys working in a collaborative, fast-paced and quickly changing environment.
